  • Dr. Meinolf Brackhagen, Senior TS&D Scientist, presentation at 09:30 on April 24 in room B entitled: Expanded Design Space in Hot Melt Extrusion with High Productivity Hypromellose Acetate Succinate.

    To date, there are few excipients that have been specifically developed for spray drying and hot melt extrusion applications. The excipients used in this application space were typically developed for other applications such as enteric coatings and are not optimized for the spray drying or hot melt extrusion process. The presentation will covers results from a study comparing AFFINISOL™ High Productivity HPMCAS, a new grade of HPMCAS designed specifically to improve processability in spray drying and hot melt extrusion, to standard (commercially available) HPMCAS. The High Productivity HPMCAS has reduced melt viscosity compared to standard commercial HPMCAS, resulting in lower die pressure and reduced torque and free acid generation. It was found that the HPMCAS polymers were easily extrudable at temperatures above 150°C (standard HPMCAS) and 135°C (HP HPMCAS). Minimized generation of free acids was observed at moderately high temperatures (150-170°C) under low shear/low residence time conditions (150-200 rpm, 400-500 g/h). It was as well found that under same conditions, HP HPMCAS allowed for extrusion with significantly lower die pressure and considerably lower drive load and free acid levels.
